To a stirred solution of 4-((2-chloro-5-nitropyrimidin-4-yl)amino)cyclohexane-1-carboxamide (0.5 g, 2.2 mmol) in DMF (10 mL) was added 2,2-dimethyltetrahydro-2H-pyran-4-amine (0.24 g, 1.8 mmol) and sodium carbonate (0.53 g, 5.0 mmol) at ambient temperature. The reaction mixture was stirred at ambient temperature for 16 h. Completion of the reaction was confirmed by UPLC. The product was isolated and triturated with petroleum ether to afford (1S,4S)-4-((2-((2,2-dimethyltetrahydro-2H-pyran-4-yl)amino)-5-nitropyrimidin-4-yl)amino)cyclohexane-1-carboxamide (0.4 g, 61%) as an yellow solid.
