Here, we report on the first combined one-pot use of the two so-called “click reactions”: The thiol-ene coupling and the copper-catalyzed alkyne-azide cycloaddition. A Sugar-Based Gelator for Marine Oil-Spill Recovery

Marine oil spills constitute an environmental disaster with severe adverse effects on the economy and ecosystem. Phase-selective organogelators (PSOGs), molecules that can congeal oil selectively from oil?water mixtures, have been proposed to be useful for oil-spill recovery. However, a major drawback lies in the mode of application of the PSOG to an oil spill spread over a large area. The proposed method of using carrier solvents is impractical for various reasons. Direct application of the PSOG as a solid, although it would be ideal, is unknown, presumably owing to poor dispersion of the solid through the oil. We have designed five cheap and easy-to-make glucose-derived PSOGs that disperse in the oil phase uniformly when applied as a fine powder. These gelators were shown to selectively congeal many oils, including crude oil, from oil?water mixtures to form stable gels, which is an essential property for efficient oil-spill recovery. We have demonstrated that these PSOGs can be applied aerially as a solid powder onto a mixture of crude oil and sea water and the congealed oil can then be scooped out. Our innovative mode of application and low cost of the PSOG offers a practical solution to oil-spill recovery.

A puzzling inversion of enantioselectivity dependent on catalyst loading was observed during the quinine-mediated desymmetrization of glutaric meso-anhydrides. This study presents the improvement of the catalytic path by the inclusion of carboxylic acid additives up to synthetically useful levels. The novel protocol utilizing 0.1 equiv of alkaloid and xanthene-9-carboxylic acid at room temperature (rt) was found comparable to the protocol requiring 1.1 equiv of alkaloid at -30 C. Thus, by altering the protocol the same catalyst produces the opposite enantiomer. This occurrence was rationalized by an extensive computational study of the interactions governing the molecular complexes formed by quinine, methanol, 3-methylglutaric anhydride, and the acetic acid. It was found that in a quinine catalyzed reaction the alcohol and the anhydride were directly hydrogen bonded to the catalyst. On the other hand, in the reaction with additive the acid intercalates between the alcohol and quinine. Due to this insertion the alcohol approaches the anhydride from the opposite face, in agreement with the observed inversion of enantioselectivity

Trehalose dimycolates (TDMs) are the most abundant glycolipids found in the cell wall of Mycobacterium tuberculosis (M. tb). TDMs play an important role in the pathogenesis of M. tb yet the only known receptor for TDM is the macrophage inducible C-type lectin (mincle). To understand more about the interaction of TDMs with immune cells, affinity based proteome profiling (AfBPP) can be used to determine receptors that bind TDMs. To this end, we present the synthesis of the first AfBPP-TDM probe and report on its ability to activate macrophages. By doing so, we establish that the AfBPP-TDM probe appears to be a suitable substrate for future proteomic profiling experiments.

Core-sheath nanostructured chitosan-based nonwovens as a potential drug delivery system for periodontitis treatment

Core-sheath nanofibers were successfully prepared via coaxial electrospinning by using chitosan with well-defined structural characteristics as the shell layer and poly (vinyl alcohol) (PVA) containing tetracycline hydrochloride (TH) as the core layer. The effects of the average degree of deacetylation (DD?) of chitosan and the post-electrospinning genipin crosslinking on physicochemical and biological properties of resulting nonwovens were evaluated. Defect-free and geometrically uniform nanofibers with diameters predominantly in the range of 100?300 nm were prepared, and transmission electron microscopy (TEM) revealed the core-sheath structures and its preservation after crosslinking. The mechanical properties, as well as the stability of nonwovens in aqueous medium, were greatly improved by genipin-crosslinking, which enabled a sustained release of TH over 14 days. Results also revealed that the release profile of TH in the presence of lysozyme was affected by the composition of the shell layer, as the TH release rate increases with decreasing of DD?. Further in vitro antimicrobial activity demonstrated that the cross-linked nonwovens containing TH showed strong activity against bacterial strains associated with periodontal disease. Additionally, the nonwovens did not demonstrate cytotoxic toward fibroblast (HDFn) cells, hence showing their potential for applications as a novel drug delivery platform for periodontitis treatment.

The synthesis of a new series of 4-acylaminopyrazolo[3,4-d]pyrimidines active on the sigma-1 receptor (sigma1R) is reported. Compounds were efficiently prepared using a two to three step process starting from commercially available 1H-pyrazolo[3,4-d]pyrimidin-4-amine. A SAR study shows that the sigma1R requires the presence of relatively highly lipophilic substituents at opposite sides of the central scaffold, while selectivity versus the sigma2R can be improved by shortening the distance of the basic nitrogen to it. Compound 9a was among the most active and selective in vitro derivatives and exhibited potent antinociceptive properties in several pain models in mice, indicating its antagonistic behaviour.
