A new type of crown compound (1-10) featuring a spiro-linked assembly of two to four individual macrorings differing in ring size, rigidity, and donor characteristics is synthesized.Syntheses were achieved by means of high-dilution techniques and blocking/deblocking procedures beginning with pentaerythritol as the basic building block.The ability of the new ligands (“multiloop crown compounds”) to selectively form crystalline polyhomonuclear/heteronuclear complexes is studied.It is found that the symmetrically looped molecules (1a-c, 4a, 6, 7d) can incorporate two orthree identical metal cations well matched in their diameter (Li+, Na+, K+, Ca2+, Ba2+).Cations which are too large to fit into the crown rings of 1a-c complex in a sandwich-like manner; those which are too small effect a high hydration in their complexes.Asymmetrical 1d and 9 allow the common uptake of different metal ions, e.g., a combination of Li+ and Ba2+ or of 2K+ and Co2+.The specific occupation of the different subrings is discussed.Ligand 1b is shown to discriminate Ca2+ from Na+; 1c and 6 preferentially complex Ba2+ from a Ba2+/K+ mixture.Thus, the new ligands act as manifold selective cation receptors, corresponding to the specification of their binding compartments.

Discovery and SAR of methylated tetrahydropyranyl derivatives as inhibitors of isoprenylcysteine carboxyl methyltransferase (ICMT)

A series of tetrahydropyranyl (THP) derivatives has been developed as potent inhibitors of isoprenylcysteine carboxyl methyltransferase (ICMT) for use as anticancer agents. Structural modification of the submicromolar hit compound 3 led to the potent 3-methoxy substituted analogue 27. Further SAR development around the THP ring resulted in an additional 10-fold increase in potency, exemplified by analogue 75 with an IC50 of 1.3 nM. Active and potent compounds demonstrated a dose-dependent increase in Ras cytosolic protein. Potent ICMT inhibitors also reduced cell viability in several cancer cell lines with growth inhibition (GI50) values ranging from 0.3 to >100 muM. However, none of the cellular effects observed using ICMT inhibitors were as pronounced as those resulting from a farnesyltransferase inhibitor.

Topological diversification in metal-organic frameworks: Secondary ligand and metal effects

A series of interesting coordination polymers have been prepared by the combination of a V-shaped 4,4?-oxybis(benzoic acid) (H2oba) and neutral organonitrogen ligands with different metal ions, namely, {[M(oba)(bpe)]¡¤H2O}n [M = Mn (1), Co (2)], [M(oba)(N3)]n [N3 = dipyridin-2-ylamine; M = Cd (3), Cu (4)] and {[Zn(oba)(N5)]¡¤2H2O}n (5) [bpe = 1,2-bis(4-pyridyl)ethene, N5 = bis(pyridin-2-yl)pyridine-2,6-diamine]. The framework structures of these neutral polymeric complexes have been determined by single-crystal X-ray diffraction studies. Compound 1 has a threefold pcu-type network structure. Although 2 has the same ligands and coordination modes as 1, it consists of corrugated 2D layers formed of a series of squares, which allows the sheets to interpenetrate in an unusual 2D?3D parallel fashion. Polymers 3 and 4 exhibit double-helical chains formed by pi-pi stacking interactions from the phenyl rings of the N3 ligands. Compound 5 forms a 2D supramolecular architecture directed by hydrogen bonding between the NH groups of the N5 ligand, the uncoordinated carboxylate oxygens, and the intercalated water molecules. This work markedly indicates that the effect of auxiliary ligands is significant in the construction of these network structures, which are also well regulated by the metal centers. Thermogravimetric analysis (TGA) and XRPD results for compound 1 as well as luminescent properties for compounds 3 and 5 are discussed. Wiley-VCH Verlag GmbH & Co. KGaA, 2009.

We report a new reagent for the functionalization of unprotected oligosaccharides with a picolyl azide group at the anomeric position for chelation-assisted copper-catalyzed alkyne-azide cycloaddition (CuAAC) glycoconjugation. We show that oligosaccharides functionalized with this moiety react with an apparent second-order rate constant of 193 m-1 s-1 and can be used to functionalize biomolecules bearing alkyne moieties introduced through metabolic labeling, including in live cells.

A new class of analogues of the bifunctional radiosensitizer alpha-(1-aziridinylmethyl)-2-nitro-1H-imidazole-1-ethanol (RSU 1069): The cycloalkylaziridines

A series of compounds related to alpha-(1-aziridinylmethyl)-2-nitro-1H-imidazole-1-ethanol (RSU 1069, 1) were synthesized and evaluated as selective hypoxic cell cytotoxic agents and as radiosensitizers. The aziridine moiety was replaced with a number of other potential alkylating groups including cycloalkylaziridines and azetidines. The data indicated that modification of the aziridine of 1 resulted in a substantial decrease in the ability of the compounds to selectively kill hypoxic cells. However, these modifications did not affect the compounds’ in vitro radiosensitizing activity since many of the derivatives were as potent as 1. All of the compounds that were evaluated in vivo were less toxic than 1, and several members of this series had significant activity. The best compound was trans-alpha-[[(4-bromotetrahydro-2H-pyran-3-yl)amino]methyl]-2-nitro-1H- imidazole-1-ethanol (18), which, due to its activity and log P value, is a candidate for additional in vivo studies.

Novel examples of hybrid organic-inorganic solids based on V4 O124 – building blocks

New hybrid organic/inorganic copper oxovanadium compounds of composition [Cu(dpa)2]4V4O12 (1), Cu2(bipy)2V4O12 (2), and Cu2(bipy)2V6O17 (3), which crystallize in the monoclinic 1, 2 and triclinic 3 symmetry, respectively, were obtained by soft hydrothermal routes. The first two structures present isolated [V4O12]4- anionic units, the structure of 3 exhibits the same [V4O12]4- cycles coupled by V2O5 dimeric bridges propagated as an infinite chain along the c-direction 3. For compound 1, the copper(I) centers are bonded to four nitrogen atoms with a distorted tetrahedral geometry. For compounds 2 and 3, the copper(II) centers are square pyramidal with two nitrogen atoms from the neutral diimine ligand and three oxygen atoms from the oxovanadium sub-unit. In all the compounds described herein, the vanadium ions are as V(V), occupying solely tetrahedral sites.
