New triterpene glycosides, ulososides C, (20S,22S,23R,24S)-3beta ,22,23-trihydroxy-3-O-(beta-D-glucopyranosyl)-32-nor-24-methyllanost-8(9) -ene-30-oic acid, D, (20S,22S,23R,24S)-3beta,22,23-trihydroxy-3-O-(beta -D-N-acetylglucosaminopyranosyl)-32-nor-24-methyllanost-8(9)-ene-30-oic acid, and E, (20S,22S,23R,24S)-3beta,22,23-trihydroxy-3-O-(beta -D-glucuronopyranosyl-(1 ? 2)-alpha -D-arabinopyranosyl-32-nor-24-methyllanost-8(9)-ene-30-oic acid, were isolated from an Ulosa sp. sponge. Their structures were determined by spectral methods and chemical transformations. Specific features of their structures are discussed.

Enzymatic synthesis of trideuterated sialosides

Sialic acids are a family of acidic monosaccharides often found on the termini of cell surface proteins or lipid glycoconjugates of higher animals. Herein we describe the enzymatic synthesis of the two isotopically labeled sialic acid derivatives d3-X-Gal-a-2,3-Neu5Ac and d3-X-Gal-a-2,3-Neu5Gc. Using deuterium oxide as the reaction solvent, deuterium atoms could be successfully introduced during the enzymatic epimerization and aldol addition reactions when the sialosides were generated. NMR and mass spectrometric analyses confirmed that the resulting sialosides were indeed tri-deuterated. These compounds may be of interest as internal standards in liquid chromatography/mass spectrometric assays for biochemical or clinical studies of sialic acids. This was further exemplified by the use of this tri-deuterated sialosides as internal standards for the quantification of sialic acids in meat and egg samples.

Synthesis and investigation of charge transport properties in adducts of hole transporting carbazole derivatives and push-pull azobenzenes

In order to investigate the viability of a material design for bulk heterojunction (BHJ) organic solar cells, where hole transporting group is bound to the donor moiety, we report the synthesis and charge transport characteristics of 3-(diphenylamino)carbazolyl-functionalized derivatives of 2-(4-((4-(dimethylamino)phenyl)diazenyl)benzylidene)-1H-indene-1,3-dione (DMAAzi) chromophore. Three different bounding configurations were examined in these adducts. Additionally, a trityl-functionalized derivative of DMAAzi was prepared and used for comparison purposes. All of the synthesized materials form thin amorphous films from volatile organic solvents and exhibit glass transition temperatures in the range from 89 C to 124 C. The molecular ionization energy and electron affinity energy levels in thin films were measured. Photo-induced time of flight (ToF) method was used in to determine charge carrier drift mobilities. It was found out that the formation of deep charge trap states with local energies at approximately 0.60?0.78 eV takes place and has a considerable negative effect on the hole drift mobility of the investigated compounds.

Aryl 1,4-diazepane compounds as potent and selective CB2 agonists: Optimization of drug-like properties and target independent parameters

A high throughput screening campaign identified aryl 1,4-diazepane compounds as potent and selective cannabinoid receptor 2 agonists as compared to cannabinoid receptor 1. This class of compounds suffered from poor drug-like parameters as well as low microsomal stability and poor solubility. Structure-activity relationships are described with a focus on improving the drug-like parameters resulting in compounds with improved solubility and permeability.

The synthesis is described of four pentasaccharides, which are structurally related to the bacterial capsular polysaccharide isolated from E. coli K5 (010/K5/H40), i.e. the so-called K5 antigen.These four synthetic compounds comprise: (i) a pentasaccharide that is structurally identical to the K5 antigen (i.e. compound 16); (ii, iii) two pentasaccharides containing two and three O-sulphated groups respectively on defined positions (i.e. compounds 15a and 15b); (iv) a pentasaccharide that is O-sulphated on all hydroxyl groups (i.e. compound 17).These four K5-antigen-related pentasaccharides were synthesized from the fully protected pentasaccharides 13a and 13b.The preparation of compounds 13a,b was based on coupling of disaccharide 6a with the disaccharide 10a or 10b to give tetrasaccharides 11a,c, respectively, which in turn were coupled to monosaccharide 12.Deblocking of the pentasaccharides 13a,b and conversion into the pentasaccharides 15a and 15b was performed by saponification of the ester functions, O-sulphation, hydrogenalysis and finally selective N-acetylation. compound 16 was obtained from 13a,b by saponification of the ester functions, deblocking of the benzyl and N-protective groups, followed by selective N-acetylation.The persulphated derivative 17 was obtained from 15a by extensive O-sulphation.The structure of the K5-antigen-related pentamers was confirmed by 1H NMR- and 13C NMR-spectroscopical techniques.

Dependency of the regio- and stereoselectivity of intramolecular, ring-closing glycosylations upon the ring size

Phenyl 3,4,6-tri-O-benzyl-2-O-(3-carboxypropionyl)-1-thio-beta-D- galactopyranoside (1) was condensed via its pentafluorophenyl ester 2 with 5-aminopentyl (4a), 4-aminobutyl (4b), 3-aminopropyl (4c) and 2-aminoethyl 4,6-O-benzylidene-beta-D-glucopyranoside (4d), prepared from the corresponding N-Cbz protected glucosides 3a-d, to give the corresponding 2-[3- (alkylcarbamoyl)propionyl] tethered saccharides 5a-d. Intramolecular, ring closing glycosylation of the saccharides with NIS and TMSOTf afforded the tethered beta(1?3) linked disaccharides 6a-c, the a(1?3) linked disaccharides 7a-d and the a(1?2) linked disaccharide 8d in ratios depending upon the ring size formed during glycosylation. No beta(1?2) linked disaccharides were formed. Molecular modeling of saccharides 6-8 revealed that a strong aromatic stacking interaction between the aromatic parts of the benzyl and benzylidene protecting groups in the galactosyl and glucosyl moieties was mainly responsible for the observed regioselectivity and anomeric selectivity of the ring-closing glycosylation step.

N-Heterocyclic Carbene Ligand-Enabled C(sp3)-H Arylation of Piperidine and Tetrahydropyran Derivatives

PdII-catalyzed C(sp3)-H arylation of saturated heterocycles with a wide range of aryl iodides is enabled by an N-heterocyclic carbene (NHC) ligand. A C(sp3)-H insertion step by the PdII/NHC complex in the absence of ArI is demonstrated experimentally for the first time. Experimental data suggests that the previously established NHC-mediated Pd0/PdII catalytic manifold does not operate in this reaction. This transformation provides a new approach for diversifying pharmaceutically relevant piperidine and tetrahydropyran ring systems. PdII-catalyzed C(sp3)-H arylation of saturated heterocycles with a wide range of aryl iodides is enabled by an N-heterocyclic carbene (NHC) ligand. A C-H insertion step by the PdII/NHC complex in the absence of ArI is demonstrated experimentally for the first time. This arylation method provides a new approach for diversifying pharmaceutically relevant piperidine and tetrahydropyran ring systems (see scheme).
