Structure, antimicrobial activity, albumin- and DNA-binding of manganese(II)-sparfloxacinato complexes

Manganese(II) complexes with the quinolone antimicrobial agent sparfloxacin (Hsf) in the absence or presence of the nitrogen-donor heterocyclic ligands 1,10-phenanthroline (phen), 2,2?-bipyridine (bipy), 2,2?-bipyridylamine (bipyam) or pyridine (py) were synthesized and characterized with diverse physicochemical and spectroscopic techniques. The crystal structure of complex [Mn(sf)2(phen)]·4MeOH was determined by X-ray crystallography. In the resultant complexes, the deprotonated sparfloxacinato ligands are bidentately bound to manganese(ii) through the pyridone oxygen and a carboxylato oxygen. The antimicrobial activity of the complexes was tested against four different microorganisms (Escherichia coli, Xanthomonas campestris, Staphylococcus aureus and Bacillus subtilis) and was found to be similar to or higher than free Hsf. The binding of the complexes to calf-thymus DNA (CT DNA) was monitored by UV spectroscopy and DNA viscosity measurements, which indicated intercalation as the most possible mode, and the DNA-binding constants of the complexes were calculated. The ability of the complexes to displace ethidium bromide (EB) from the EB-DNA complex was also investigated. Fluorescence emission spectroscopy was used to evaluate the interaction of the complexes with human or bovine serum albumin proteins revealing their binding with relatively high binding constant values. The synthesis of sialic acids and their analogs was accomplished based on substrate-controlled asymmetric aldol reactions between sterically complicated aldehydes easily prepared from commercially available carbohydrates and a novel pyruvic acid oxabicyclo[2.2.2]octyl orthoester. Systematic aldol reaction studies using chiral aldehydes revealed that alpha,beta,gamma-benzyloxy-substituted aldehydes with an alpha,beta-anti relative configuration preferentially provided the Felkin products with the 4,5-anti configuration with high diastereoselectivity. The relative beta,gamma-configuration in alpha,beta,gamma-benzyloxy-substituted aldehydes with an alpha,beta-syn arrangement exerted a secondary effect on the diastereoselectivity of the stereogenic center formed in aldol reactions, and alpha,beta-syn-beta,gamma-anti benzyloxyaldehyde exhibited superior diastereoselectivity to alpha,beta-syn-beta,gamma-syn benzyloxyaldehyde to yield the Felkin products.

beta-Glucuronyl carbamate based pro-moieties designed for prodrugs in ADEPT

A number of pro-moieties 8a-e designed for prodrug preparation have been synthesized. The pro-moieties, containing a glucuronyl carbamate group linked to a spacer possessing a terminal carboxylic acid group, have been synthesized from isocyanates 6 and anomerically unprotected glucuronic acids 10. The requisite isocyanates had to be prepared using the Curtius rearrangement. Glucuronyl carbamates proved to be excellent substrates for human beta-glucuronidase. The pro-moieties 8a-e can be coupled to hydroxy- or amino group containing drugs. The resulting prodrugs are designed to be activated by beta-glucuronidase and to be used in ADEPT. Application is demonstrated with the synthesis of duanomycin prodrugs 12a-e.

This core was combined with elements of screening hits 2, 19, and 33 and resulted in highly potent, selective tankyrase inhibitors that are novel three pocket binders. NVP-TNKS656 (43) was identified as an orally active antagonist of Wnt pathway activity in the MMTV-Wnt1 mouse xenograft model. With an enthalpy-driven thermodynamic signature of binding, highly favorable physicochemical properties, and high lipophilic efficiency, NVP-TNKS656 is a novel tankyrase inhibitor that is well suited for further in vivo validation studies.

Synthesis and aggregation behaviour of single-chain, 1,32-alkyl branched bis(phosphocholines): Effect of lateral chain length

Three novel single-chain bis(phosphocholines) bearing two lateral alkyl chains of variable length next to the headgroup have been synthesized as model lipids for naturally occurring archaeal membrane lipids. The synthesis was realized using the Cu-catalyzed Grignard bis-coupling reaction of a primary bromide as a side part and a 1,omega-dibromide as a centre part. We could show that the aggregation behaviour of the resulting bolalipids strongly depends on the length of the lateral alkyl chain: the C3-branched bolalipid self-assembles into lamellar sheets, whereas the C6- and C9-analogues form nanofibres. The lamella-forming bolalipids could be used in the future to prepare stable and tailored liposomes for oral drug delivery.

Two approaches to the total synthesis of (+/-)-trichodiene (1) are reported from cis-fused lactone 10.Sequential alkylation of lactone 10 was observed to be highly stereoselective as established by the construction of diastereomeric lactone esters 12 and 14.Lactone ester 12 was utilized to prepare (+/-)-trichodiene (1) with a high degree of regioselectivity.All attempts to synthesize (+/-)-bazzanene (2), the diastereomer of (+/-)-trichodiene (1), from lactone 13 were found to be unsuccessful in the last stage of the synthetic scheme.
