Lower rim 1,3-di-amide-derivative of calix[4]arene possessing bis-{N-(2,2?-dipyridylamide)} pendants: a dual fluorescence sensor for Zn2+ and Ni2+

Single crystal XRD structure of the lower rim 1,3-di-amide-derivative of calix[4]arene possessing bis-{N-(2,2?-dipyridylamide)} pendants (L) exhibit two distinct binding cores, viz., N4 and O6. L was found to be selective for Zn2+ by switch-on and for Ni2+ by switch-off fluorescence by forming 1:1 complexes. The binding and the composition of the complex formed have been addressed based on steady state and time-resolved fluorescence spectroscopy in addition to the absorption and ESI MS. As L can detect Zn2+ and Ni2+ to a concentration as low as 142 and 203 ppb, respectively, L can be a very sensitive molecular probe for these ions. The coordination details of the metal ion-bound complexes have been addressed based on ab initio calculations showing that the stabilization energies are commensurate with the coordination formed.

Study of some mixed ligand complexes of tertiary diimines and bidentate amide ligands

Glycoside cleavage by a new mechanism in unsaturated glucuronyl hydrolases

Unsaturated glucuronyl hydrolases (UGLs) from GH family 88 of the CAZy classification system cleave a terminal unsaturated sugar from the oligosaccharide products released by extracellular bacterial polysaccharide lyases. This pathway, which is involved in extracellular bacterial infection, has no equivalent in mammals. A novel mechanism for UGL has previously been proposed in which the enzyme catalyzes hydration of a vinyl ether group in the substrate, with subsequent rearrangements resulting in glycosidic bond cleavage. However, clear evidence for this mechanism has been lacking. In this study, analysis of the products of UGL-catalyzed reactions in water, deuterium oxide, and dilute methanol in water, in conjunction with the demonstration that UGL rapidly cleaves thioglycosides and glycosides of inverted anomeric configuration (substrates that are resistant to hydrolysis by classical glycosidases), provides strong support for this new mechanism. A hydration-initiated process is further supported by the observed UGL-catalyzed hydration of a C-glycoside substrate analogue. Finally, the observation of a small beta-secondary kinetic isotope effect suggests a transition state with oxocarbenium ion character, in which the hydrogen at carbon 4 adopts an axial geometry. Taken together, these observations validate the novel vinyl ether hydration mechanism and are inconsistent with either inverting or retaining direct hydrolase mechanisms at carbon 1.

A convenient synthesis of 2-amino-3,4,6-tri-O-benzyl-2-deoxy-beta-D- glucopyranoside was described from the readily available starting material 2-acetamido-2-deoxy-D-glucose (N-acetyl-D-glucosamine). Herein, the coupling of different lipophilic amino acids with 2-amino-3,4,6-tri-O-benzyl-2-deoxy-beta- D-glucose was reported via an amide linkage as useful building blocks for the synthesis of glycopeptides. Of particular interest, bioactive peptide Arg-Gly-Asp (RGD) was incorporated into the building block containing valine was also reported. The 15 examples of corresponding di-, tri- and tetra-peptides were obtained as single aanomers.

Manganese(II) complexes with the non-steroidal anti-inflammatory drug tolfenamic acid: Structure and biological perspectives

Manganese(II) complexes with the non-steroidal anti-inflammatory drug tolfenamic acid (Htolf) with the nitrogen-donor heterocyclic ligands 1,10-phenanthroline (phen), pyridine (py), or 2,2?-bipyridylamine (bipyam) and/or the oxygen-donor ligands H2O or N,N-dimethylformamide (DMF) have been synthesized and characterized. The crystal structures of complexes [Mn(tolf-O)(tolf-O,O?)(phen)(H2O)], [Mn2(mu 2-tolf-O,O?)2(tolf-O,O?)2(bipyam) 2], [Mn2(mu2-H2O) (mu2-tolf-O,O?)2(tolf-O)2(py) 4]·1.5MeOH·py, and [Mn(mu2-tolf-O, O?)2(DMF)2]n have been determined by X-ray crystallography. The interaction of the complexes with serum albumin proteins was investigated, and relative high binding constant values were calculated. The ability of the compounds to scavenge 1,1-diphenyl- picrylhydrazyl, 2,2?-azinobis(3-ethylbenzothiazoline-6-sulfonic acid), and hydroxyl radicals was evaluated, and [Mn(tolf)2(phen)(H 2O)] was the most active scavenger among the compounds. The compounds have also exhibited noteworthy in vitro inhibitory activity against soybean lipoxygenase. UV titration studies of the interaction of the complexes with calf-thymus (CT) DNA have proved the binding to CT DNA with [Mn(mu 2-tolf)2(DMF)2]n exhibiting the highest DNA-binding constant (Kb = 5.21 (±0.35) × 105 M-1). The complexes bind to CT DNA probably via intercalation as suggested by DNA-viscosity measurements and competitive studies with ethidium bromide (EB), which revealed the ability of the complexes to displace the DNA-bound EB.

An Efficient Way to 1,7-Enynes and Ethyl 8-Yn-2-enoates from Aldohexoses and to Polyhydroxylated 1-Vinylcyclohexenes

In this paper, we report the synthesis of carbohydrate-derived 1,7-enynes and subsequent metathesis to yield polyhydroxylated 1-vinylcyclohexenes. For example, we converted D-glucose 2 to the (6,7)-dideoxy-D-gluco-hept-6-ene-pyranose 7, which led to the desired 1,7-enyne 16. The ring-closing metathesis of this 1,7-enyne 16 with the second generation Grubbs catalyst, under Mori’s conditions, gave the corresponding polyhydroxylated 1-vinylcyclohexene 25 in 76% yield. The conversion of several aldohexoses into polyhydroxylated 1-vinylcyclohexenes was carried out with satisfying yields. We report also the synthesis of two carbohydrate-derived ethyl 8-yn-2-enoates from D-glucose derivatives.

Multi-component synthesis of peptide-sugar conjugates

Recent years have witnessed a growing interest in the development of new methods for linking sugars to peptides or proteins because natural glycopeptides or neoglycoconjugates with well defined chemical structures are very important tools to study diverse biological phenomena. Herein we report a novel, one-pot, three-component process for the synthesis of peptide-urea conjugates incorporating a hexafluorovaline or an aspartic acid alkyl ester residue under very mild conditions and high yields. The reaction has been exploited for the synthesis of a wide array of structurally diverse peptide-sugar conjugates through a regiospecific four-component, one-pot sequential domino process, by generating the reacting sugar-carbodiimides in situ from readily accessible starting materials.
