A convenient synthesis of 2-amino-3,4,6-tri-O-benzyl-2-deoxy-beta-D- glucopyranoside was described from the readily available starting material 2-acetamido-2-deoxy-D-glucose (N-acetyl-D-glucosamine). Herein, the coupling of different lipophilic amino acids with 2-amino-3,4,6-tri-O-benzyl-2-deoxy-beta- D-glucose was reported via an amide linkage as useful building blocks for the synthesis of glycopeptides. Of particular interest, bioactive peptide Arg-Gly-Asp (RGD) was incorporated into the building block containing valine was also reported. The effects of organic solvents on the synthesis of galactose disaccharides using beta-galactosidases

The use of beta-galactosidases from five different sources in syntheses through transglycosylation leads to 3-O-methyl allolactose, N-acetyllactosamine, N-acetylallolactosamine and Gal(beta1-6)Gal. The yield and distribution of products depends on the source of the enzyme and on the reaction conditions, viz. the nature of added organic co-solvent. Yields of 3-O-methyl allolactose (47%), Gal(beta1-6)Gal (6%), N-acetylallolactosamine (30%) were obtained in aqueous buffer with beta-galactosidase from E. coli. The same reactions occurred at much lower rates in the presence of water-miscible organic solvents. With beta-galactosidases from K. fragilis and A. oryzae, however, the synthesis of the above disaccharides occurred only in the presence of organic solvent (> 60% v/v triethyl phosphate, trimethyl phosphate or tetraglyme) but not in aqueous buffer solution. beta-galactosidases from D. pneumoniae and B. circulans in systems incorporating organic solvent produced 3-O-methyl allolactose and N-acetyllactosamine in yields of 30-40%. The direct separation of oligosaccharides on a preparative scale can be achieved by Ca2+-ligand exchange chromatography. Ultrafiltration was also used for the efficient recycling of enzymes.

The conversion of sugars into glycomimetics typically involves multiple protecting-group manipulations. The development of methodology allowing the direct aqueous conversion of free sugars into glycosides, and mimics of oligosaccharides and glycoconjugates in a high-yielding and stereoselective process is highly desirable. The combined use of 2-azido-1,3-dimethylimidazolinium hexafluorophosphate and the Cu-catalyzed Huisgen cycloaddition allowed the synthesis of a range of glycoconjugates in a one-step reaction directly from reducing sugars under aqueous conditions. The reaction, which is completely stereoselective, may be applied to the convergent synthesis of triazole-linked glycosides, oligosaccharides, and glycopeptides. The procedure provides a method for the one-pot aqueous ligation of oligosaccharides and peptides bearing alkyne side chains.

Kinetics of hydrolysis of chitin/chitosan oligomers in concentrated hydrochloric acid

The kinetics of hydrolysis in concentrated hydrochloric acid (12.07 M) of the fully N-acetylated chitin tetramer (GlcNAc4) and the fully N-deacetylated chitosan tetramer (GlcN4) were followed by determining the amounts of the lower DP oligomers as a function of time. A theoretical model was developed to simulate the kinetics of hydrolysis of the three different glycosidic linkages in the tetramers. The model uses two different rate constants for the hydrolysis of the glycosidic bonds in the oligomers, assuming that the glycosidic bond next to one of the end residues are hydrolysed faster than the two other glycosidic linkages. The two rate constants were estimated by fitting model data to experimental results. The results show that the hydrolysis of the tetramers is a nonrandom process as the glycosidic bonds next to one of the end residues are hydrolysed 2.5 and 2.0 times faster as compared to the other glycosidic linkages in the fully N-acetylated and fully N-deacetylated tetramer, respectively. From previous results on other oligomers and the reaction mechanism, it is likely that the glycosidic bond that is hydrolysed fastest is the one next to the nonreducing end. The absolute values for the rate constants for the hydrolysis of the glycosidic linkages in GlcNAc4 were found to be 50 times higher as compared to the glycosidic linkages in GlcN4, due to the catalytic role of the N-acetyl group and the presence of the positively charged amino-group on the N-deacetylated sugar residue.

Synthesis and antibacterial activity of doxycycline neoglycosides

A set of 37 doxycycline neoglycosides were prepared, mediated via a C-9 alkoxyamino-glycyl-based spacer reminiscent of that of tigecycline. Subsequent in vitro antibacterial assays against representative drug-resistant Gram negative and Gram positive strains revealed a sugar-dependent activity profile and one doxycycline neoglycoside, the 2?-amino-alpha-d-glucoside conjugate, to rival that of the parent pharmacophore. In contrast, the representative tetracycline-susceptible strain E. coli 25922 was found to be relatively responsive to a range of doxycycline neoglycosides.
