Reconstituting poly(glycerol phosphate) wall teichoic acid biosynthesis in vitro using authentic substrates

Wall teichoic acids (WTAs) are phosphate-rich anionic polymers that constitute a substantial portion of the Gram-positive cell wall. Recent work has demonstrated the importance of WTAs in cell shape, virulence and antibiotic resistance. These findings highlight WTA biosynthetic enzymes as attractive targets for novel antimicrobial agents. Due to challenges involved in the isolation of natural substrates, in vitro studies of the recombinant enzymes have largely employed soluble substrate analogues. Herein we present a semisynthetic approach to obtain the authentic precursor for WTA biosynthesis, Lipid alpha, complete with its polyisoprenoid lipid moiety. We show that this material can be used to reconstitute the activities of four enzymes involved in poly(glycerol phosphate) WTA biosynthesis in a detergent micelle. This work enables the creation of chemically defined and realistic systems for the study of interfacial catalysis by WTA biosynthetic machinery, which could aid efforts to discover and develop novel agents against WTA biosynthesis.

Enzymatic synthesis of trideuterated sialosides

Sialic acids are a family of acidic monosaccharides often found on the termini of cell surface proteins or lipid glycoconjugates of higher animals. Herein we describe the enzymatic synthesis of the two isotopically labeled sialic acid derivatives d3-X-Gal-a-2,3-Neu5Ac and d3-X-Gal-a-2,3-Neu5Gc. Using deuterium oxide as the reaction solvent, deuterium atoms could be successfully introduced during the enzymatic epimerization and aldol addition reactions when the sialosides were generated. NMR and mass spectrometric analyses confirmed that the resulting sialosides were indeed tri-deuterated. These compounds may be of interest as internal standards in liquid chromatography/mass spectrometric assays for biochemical or clinical studies of sialic acids. This was further exemplified by the use of this tri-deuterated sialosides as internal standards for the quantification of sialic acids in meat and egg samples.

The synthesis of sialic acids and their analogs was accomplished based on substrate-controlled asymmetric aldol reactions between sterically complicated aldehydes easily prepared from commercially available carbohydrates and a novel pyruvic acid oxabicyclo[2.2.2]octyl orthoester. Systematic aldol reaction studies using chiral aldehydes revealed that alpha,beta,gamma-benzyloxy-substituted aldehydes with an alpha,beta-anti relative configuration preferentially provided the Felkin products with the 4,5-anti configuration with high diastereoselectivity. The relative beta,gamma-configuration in alpha,beta,gamma-benzyloxy-substituted aldehydes with an alpha,beta-syn arrangement exerted a secondary effect on the diastereoselectivity of the stereogenic center formed in aldol reactions, and alpha,beta-syn-beta,gamma-anti benzyloxyaldehyde exhibited superior diastereoselectivity to alpha,beta-syn-beta,gamma-syn benzyloxyaldehyde to yield the Felkin products.

Three different building blocks have been synthesised and used for the synthesis of linear triazole linked pseudo oligosaccharides with copper(I)-catalysed cycloaddition (CuAAC). Ethynylferrocene has been used as analytical probe to improve the UV/Vis properties and HPLC methods have been used and optimised for the analysis of the pseudo oligosaccharides. The smallest ones have been isolated and characterised by analytical HPLC, NMR, ESI-MS and elemental analysis.

The supramolecular model was further optimized by molecular dynamics simulation. Moreover, chirality successfully expressed in columnar hexagonal mesophase by dendronized carbohydrate molecules may provide inspiration in searching for chiral mesophase of carbohydrate liquid crystals.

Here we utilize tandem mass spectrometry, isotopic labeling, gas-phase hydrogen/deuterium exchange, and ion mobility measurements to characterize structures of the various product ions of protonated N-acetylhexosamines. Following the facile loss of the reducing end hydroxyl group, we identify two primary fragmentation pathways. Detailed mapping of each step in the fragmentation pathway provides new insight into the mechanisms that drive collision-induced dissociation of protonated carbohydrates. Several of the smaller fragment ions are mixtures of structural isomers, and the relative distributions of these structures reveals information about the stereochemistry of the precursor molecule.
