One-Step Derivatization of Reducing Oligosaccharides for Rapid and Live-Cell-Compatible Chelation-Assisted CuAAC Conjugation

We report a new reagent for the functionalization of unprotected oligosaccharides with a picolyl azide group at the anomeric position for chelation-assisted copper-catalyzed alkyne-azide cycloaddition (CuAAC) glycoconjugation. We show that oligosaccharides functionalized with this moiety react with an apparent second-order rate constant of 193 m-1 s-1 and can be used to functionalize biomolecules bearing alkyne moieties introduced through metabolic labeling, including in live cells.

An efficient conversion of N-acetyl-d-glucosamine to N-acetyl-d- galactosamine and derivatives

2-Acetamido-2-deoxy-d-galactose (d-GalNAc) is an important monosaccharide widely distributed in nature. However, unlike its 4-epimer, the 2-acetamido-2-deoxy-d-glucose (d-GlcNAc), d-GalNAc is very expensive to obtain from commercial sources. Herein we report an efficient transformation that allows for the conversion of d-GlcNAc to a d-GalNAc derivative 7 in three steps and in 58.4-75% overall yields. The process was carried out on a greater than 20-g scale without the need of chromatography. The versatility of compound 7 was demonstrated by the synthesis of several useful monosaccharides and thiodisaccharides containing a d-GalNAc residue.

We describe a general method that permits characterization of 100-pg to 1-ng quantities of sugars.Thus, oligosaccharides are subjected to methanolysis, followed by per-p-bromobenzoylation or pernaphthoylation, and the methyl glycoside peresters are separated by high-pressure liquid chromatography.The method is also applicable to amino and acetamido sugars.If required, the separated sugar derivatives may be unequivocally identified by the characteristic quasi-M+ peaks of bromobenzoates and extrema of excitation-split circular-dichroism (c.d.) curves.Furthermore, as the c.d. extrema of polybenzoates are predictable from extrema of component dibenzoate units, the c.d. data yield information on the unknown spatial disposition of -OH, -NH2, and -NHAc groups of glycopyranoses.Application of the method is demonstrated with viridopentaose C, a pentasaccharide obtained from the antibiotic sporaviridin.

Synthesis and characterization of d-glucosamine-derived low molecular weight gelators

Carbohydrate-based low molecular weight gelators are an interesting class of molecules with many potential applications. Previously, we have found that certain esters and carbamates of 4,6-O-benzylidene-alpha-d-methyl- glucopyranoside are low molecular weight gelators for a variety of solvents, including water. In order to obtain effective and robust sugar-based organogelators and understand the structure and gelation relationship, we extended our studies using 4,6-O-benzylidene-alpha-d-methyl-2-deoxy-2-amino- glucopyranoside as the headgroup. A series of amides and ureas were prepared from the protected d-glucosamine and the corresponding isocyanates or acid chlorides, in good yields. The self-assembling properties of these compounds were studied in several solvents, including water and aqueous solutions. Comparing to the ester and carbamate derivatives previously prepared from d-glucose, the amides and urea derivatives afforded more robust gels at lower concentrations typically. Most of these compounds were found to be efficient low molecular weight hydrogelators (LMHGs) for aqueous solutions at concentrations lower than 0.5 wt %. The preparation and characterization of these compounds are reported here.

One-pot stereoselective synthesis of beta-N-aryl-glycosides by N-glycosylation of aromatic amines: application to the synthesis of tumor-associated carbohydrate antigen building blocks

We studied the stereoselective synthesis of several beta-N-aryl-glycosides by glycosylation of aromatic primary amines using unprotected carbohydrates in aqueous solution. This was the first report showing an efficient method for the synthesis with one step of beta-N-glycosyl-para-amino-phenyl alanine building blocks for the tumor-associated carbohydrate antigen (TACA) glycopeptides synthesis. Analysis of products by 1H and 13C NMR indicated that the Amadori rearrangement had not occurred after formation of the stereoselective beta-N-glycoside bond (natural N-glycoprotein linkage). The study of the chemical and enzymatic stability in aqueous media of beta-N-aryl-glycosides synthesized was also investigated. For the first time we have shown that the N-glycosidic bond was relatively stable at pH near to 7 and more stable than the O-glycosidic bond to enzymatic hydrolysis. This higher enzymatic and chemical stability of the N-glycosidic bond is essential to envisage further development of stable TACA building blocks.
