The Pd-catalyzed intramol. addition of carbamoyl chlorides and aryl halides across alkynes is investigated by means of DFT calculations and mechanistic test experiments The data suggest a mechanistic pathway that involves oxidative addition, alkyne insertion, cis → trans isomerization and reductive elimination. Our data indicate that oxidative addition is the reactivity limiting step in the addition of aryl chlorides and bromides across alkynes. However, for the corresponding addition of carbamoyl chlorides, alkyne insertion is found to be limiting. Full energetic reaction pathways for the intramol. additions across alkynes are presented herein and the role of ligands, alkyne substituents and tether moieties are discussed. Notably, the calculations could rationalize a pronounced effect of the alkyne substituent, which accounts for the exceptional reactivity of TIPS-substituted alkynes. In particular, the bulky silyl moiety is shown to significantly destabilize the formed Pd(II)-intermediates, thus facilitating both cis → trans isomerization and reductive elimination, which overall results in a flatter energetic landscape and a therefore increased catalytic efficiency.

This work describes the chemo- and regioselective direct aminocarbonylation of alkynes withh aminophenols to form hydroxy-substituted α,β-unsaturated amides in good to excellent yields. On the other hand, branched amides could be approached by introducing 1,3-bis(diphenylphosphino)propane as the ligand and p-TsOH·H2O as the additive. In addition to the hydroxyl group, other functional substituents, such as carboxyl and vinyl groups, could also be tolerated using this method. As an application of this strategy, the natural product avenanthramide A could be synthesized directly in 84% yield and in 99% regioselectivity via the carbonylation of 2-amino-5-hydroxybenzoic acid and 4-ethynylphenol. Further studies show that the ligands and the additives are keys to good yields and selectivities.

A facile, solid-state mechanochemical method was developed for the synthesis of low molecular weight chitosan (LMWC) in one step from chitin and crude shrimp shell powders, in which chitin undergoes simultaneous deacetylation and depolymerisation in the presence of a base catalyst under mechanical milling. The method is advantageous over traditional multi-step methods, featuring enhanced efficiency and significantly reduced environmental impact. Compared to traditional approaches, the base usage is reduced to about 1/10, and the molecular weight of the obtained LMWC product is much more narrowly distributed, with a polydispersity value of only 1.1. The degree of deacetylation and the molecular weight can be adjusted by varying the ball milling parameters. The influences of different types of bases were investigated by solid-state NMR analysis and control experiments, pointing out the critical role of base in both depolymerisation and deacetylation. Base-catalysed mechanochemical transformation of chitin and shrimp shells provides a solvent-free way to effectively valorize shellfishery waste for valuable products.

Hydrogels formed by polyelectrolyte complexation (PEC) of oppositely charged biopolymers, free of any chemical additives, are promising biomaterials. In this work, the mechanical behavior of hydrogels consisting of positively charged chitosan and negatively charged sodium hyaluronate (HA) at balanced charge composition is investigated. These hydrogels exhibit strong tensile strain and strain rate dependence. They are elastic-like, independent of the strain rate at small strain, but exhibit plastic-like behavior above the yield point by showing a monotonous decrease of the stress. The cyclic tensile test demonstrates that these hydrogels exhibit small and quickly recoverable hysteresis in the elastic-like region but large and partially recoverable hysteresis above the yield point. The stress relaxation experiment shows a plateau in the reduced stress followed by an abrupt fracture, and the time to failure decreases exponentially with increasing applied step strain. Such elastic-to-plastic-like transformation of the biopolymer PEC gels is quite different from the behaviors of PEC hydrogels formed by oppositely charged vinyl-type synthetic polyelectrolytes due to the difference in flexibility, charge density, and ionic bond strength of these polymers.
