The use of ionic liquids as alternative solvents for enzyme catalysis was investigated. beta-Galactosidase from Bacillus circulans catalyses the synthesis of N-acetyllactosamine starting from lactose and N-acetylglucosamine in a transglycoslyation reaction. The addition of 25% v/v of 1,3-di-methyl-imidazol-methyl sulfate [MMIM] [MeSO4] as a water-miscible ionic liquid suppresses the secondary hydrolysis of the formed product, resulting in doubling the yield to almost 60%. The enzyme can be reused several times after ultrafiltration of the reaction mixture without loss of activity. Results of different amounts of ionic liquids in the reaction medium on the thermostability of the galactosidase as well as on oxidoreductases are presented as well.

All-cis-14,15-epoxyeicosa-5,8,11-trienoic acid (14,15-EET) is a labile, vasodilatory eicosanoid generated from arachidonic acid by cytochrome P450 epoxygenases. These data reveal differential structural parameters for the two pharmacophores that could assist the development of potent and specific in vivo drug candidates.

A lectin with insecticidal property against the stored product pest, Callosobruchus maculatus was successfully isolated from the seeds of Canavalia virosa using standard affinity chromatography. The isolated molecule typically behaved like a lectin in its characteristics. It agglutinated indicator red blood cells (RBC) in its native as well as enzyme treated conditions. The enzyme treated RBC types exhibited a very high hemagglutination (HA) titre values and this property of isolated molecule behaved like arcelin, the lectin-like molecules reported from several species of Phaseolus. As a characteristic feature of a lectin, the isolated molecule effectively inhibited the agglutination of indicator RBC types with simple and complex carbohydrates including glycoproteins. This nature of the isolated molecule also relate with characteristic feature of arcelin isoforms in inhibiting HA activity with complex glycoproteins as reported in many studies. Most interestingly, the present study disclosed trehalose as a potent inhibitor of C. virosa lectin. Therefore, feeding insect pests on the lectin like arcelin could serve as antibiosis factor/anti-insect activity. The molecular characteristics of this isolated molecule and its mass studies too revealed its homology with arcelin, arcelin-1, 2 and 6 isoforms of P. vulgaris and lectin from Canavalia cathartica, C. lineata and C. brasiliensis.

Optical, chirooptical and thermodynamic techniques have been used to study the binding between copper (II) ions and carboxyethylscleroglucan, oxidised glucomannan, 6-oxychitin and reacetylated chitosan in dilute aqueous solution. For oxidised glucomannan and reacetylated chitosan, the binding of lead was also studied. The results obtained were consistent with the occurrence of a chelate that was evidenced by the existence of a charge transfer band in the near UV region and supported by other experimental data. The variations on binding of the thermodynamic functions Gibbs free energy, enthalpy and hence entropy were computed. Indications on the site(s) of binding were derived. In the case of oxidised glucomannan, the UV spectrophotometry revealed the presence of two sites of binding, the carboxyl groups of glucuronic and mannuronic acids. Finally, the possibility of avoiding misleading results with appropriate procedures is shown.

These studies were conducted to determine whether the biosynthesis of the lipid-linked repeating tetrasaccharide was initiated by the formation of GalNAc-P-P-Und by WecA. When membrane fractions from E. coli strains K12, O157, and PR4019, a WecA-overexpressing strain, were incubated with UDP-[3H]GalNAc, neither the enzymatic synthesis of[3H]GlcNAc-P-P-Und nor [ 3H]GalNAc-P-P-Und was detected. However, when membrane fractions from strain O157 were incubated with UDP-[3H]GlcNAc, two enzymatically labeled products were observed with the chemical and chromatographic properties of [3H]GlcNAc-P-P-Und and [3H]GalNAc-P-P-Und, suggesting that strain O157 contained an epimerase capable of interconverting GlcNAc-P-P-Und and GalNAc-P-P-Und. The presence of a novel epimerase was demonstrated by showing that exogenous [3H]GlcNAc-P-P-Und was converted to [3H]GalNAc-P-P-Und when incubated with membranes from strain O157. When strain O157 was metabolically labeled with [ 3H]GlcNAc, both [3H]GlcNAc-P-P-Und and [ 3H]GalNAc-P-P-Und were detected. Transformation of E. coli strain 21546 with the Z3206 gene enabled these cells to synthesize GalNAc-P-P-Und in vivo and in vitro. The reversibility of the epimerase reaction was demonstrated by showing that [3H]GlcNAc-P-P-Und was reformed when membranes from strain O157 were incubated with exogenous [3H]GalNAc-P-P-Und. The inability of Z3206 to complement the loss of the gne gene in the expression of the Campylobacter jejuni N-glycosylation system in E. coli indicated that it does not function as a UDP-GlcNAc/UDP-GalNAc epimerase. Based on these results, GalNAc-P-P-Und is synthesized reversibly by a novel GlcNAc-P-P-Und epimerase after the formation of GlcNAc-P-P-Und by WecA in E. coli O157.
